Résumé

Le but de cette étude est de développer et de caractériser un algorithme pouvant calculer la vitesse des globules rouges à partir d'images de microcirculation sanguine in vivo. La réalisation d'un modèle in vitro et l'utilisation d'images de synthèse ont permis d'atteindre ces objectifs. Une caméra ultra rapide filme le sang qui circule à un débit connu dans le modèle in vitro. Les images de synthèse sont produites par un modèle mathématique afin de recréer les effets optiques du microscope. La corrélation croisée est utilisée conjointement à des filtres temporels et spatiaux pour calculer la vitesse des globules rouges. L'estimé du débit sanguin comparé au débit de la pompe permet de calculer la précision de l'algorithme. Les résultats montrent que la dispersion des erreurs moyennes des débits estimés est de 2,3% et que les effets optiques affectent la précision des vitesses calculées. L'algorithme pourra donc être utilisé en microscopie intravitale.

The aim of this study was to develop and evaluate the precision of an algorithm that can compute the speed of red blood cells in blood microcirculation in vivo. The creation of an in vitro model which accurately reproduced the images of the blood microcirculation and the use of virtual blood flow images enabled the achievement of these objectives.

The micro fluidic device is composed of a glass capillary, a syringe pump and an optical microscope equipped with a high-speed camera. The blood is pumped within the glass capillary and the images recorded by the camera are then post-treated by a computer. The composite images were created by a 2D projection of a three-dimensional mathematical blood microcirculation model. They were created expressly to reproduce the optical effects produced by the microscope. The algorithm used to calculate the speed of the red blood cells is the cross-correlation. The size of the first and second interrogation windows were optimized for the images of blood flow. The spurious speed vectors were filtered by temporal and spatial filters.

The flow estimate from the speed of red blood cells was compared to the pump flow to validate the precision of the speed algorithm. The analysis of results shows a mean error dispersion of 2.3% for physiological flows between 1 and 27 uL/h. Thus, the algorithm can be used on intravital microscopic images since the precision of the red blood cell speed is assured.
